The end-user provided virus (e.g. as a tick homogenate) will be used to experimentally infect castor bean tick nymphs (Ixodes ricinus). The nymphs will be infected by micro-capillary inoculation into the coxal plate of the second pair of legs (transcoxal infection) using a digital microinjector system. First, 100 nymphs will be used for evaluation of the virus replication dynamics. Based on the obtained results, tick-derived virus stock will be generated from an additional 100 nymphs and provided to the end-user.

The access will comprise a detailed theoretical and subject to availability practical application of the biosafety and biosecurity management at Institut Pasteur. Among others, establishment and validation of suitable decontamination methods for waist, rooms and material, as well as documentation and information management will be addressed.

Institut Pasteur Paris - General description of fluorecent antibody tets (FAT), virus isolation, lateral flow device in the context of rabies diagnosis and surveillance. Practical training in these technique in BSL3.

Institut Pasteur Paris - In this 2 weeks course the basic principles of routine virus typing will be addressed by a combination of lectures, demonstration and hands-on training. Participants will perform all required laboratory and analysis procedures for routine typing of lyssaviruses: Special attention will be given to quality control and assay validation.
