Product Description
Ref-SKU:
007N-05798 full length plasmid clone of proviral HIV-1; construct produces defective HIV-1 upon transfection into eukaryotic cells (frame-shift in gag); based on NL-BX

Special feature:
Can it be used to produce GMO:
No
Biosafety restrictions:
Upon transfection of eukaryotic cells, DNA produces viral proteins lacking functional Gag; does not produce infectious particles.

Mutations:
Contain mutations (no frameshift, no unexpected STOP codon)
Observed mutations:
The virus genome is derived from pNL-NF (pNL4-3 with shortened cellular flank) (doi.org/10.1016/j.jcv.2006.05.001).
mutations as indicated in the name; backbone is NL-BX.
A unique BamHI site was introduced 5’ of protease (ggagcc2207ggaTcc; A->S in Pol; E->D in Gag) and an XmaI site 3’ of protease (ccagga2589ccCggG).
The original BamHI site as position 8465 was removed (GGATCC->GGATCA change silent in Env; L->S in Rev).
clone has a C deletion at position 2101 => frameshift in gag, rendering the virus defective!

Note:
a virus-inactivating frameshift (one nucleotide deletion) is present in the coding sequence of gag: C deletion at position 2101);
